Human Leukocyte Antigen Class III (HLA Class III) holds genetic polymorphisms that correlate with the etiopathogenesis, susceptibility and severity of diseases that affect humans. Objective: The study aims to clarify the role of polymorphisms and the actions present in HLA Class III in the symptoms and pathogenesis of chronic, autoimmune, inflammatory, carcinogenic and infectious diseases that affect humanity. Methodology: This is an Integrative Review, based on the acronym PCC and including the databases Virtual Health Library (VHL), Capes Periódicos, Google Scholar, Embase, Medical Literature Analysis and Retrieval System Online (MEDLINE), Brazilian Bibliography of Dentistry (BBO), ScienceDirect and Scopus. Terms of interest found in the Medical Subject Headings (MeSH) and in the Health Sciences Descriptors (DeCS) were used as descriptors. Results: 56 studies were retrieved, including 35 case-control articles, 3 in vivo study articles, 2 literature review articles, 15 cohort study articles and 1 case report article. HLA Class III polymorphisms are due to isolated and coordinated mutations with other gene loci. Conclusion: HLA Class III polymorphisms can directly or indirectly corroborate diseases that affect some human populations.
