Introduction: Insomnia is a prevalent and complex sleep disorder that significantly affects the quality of life and physical and mental well-being of individuals. Objective: Explore the pathophysiological bases of insomnia and discuss multidisciplinary treatment strategies. Methods: This is a bibliographic review, using articles present in the following databases: PubMed, Web of Science, Scopus and Google Scholar. Original articles and systematic reviews in English and Portuguese published between 2009 and 2024 were included, which addressed the pathophysiological bases and multidisciplinary treatment of insomnia. After analysis, 18 articles were selected to prepare this bibliographic review. Results and Discussion: The neurobiological bases of insomnia are complex and involve an intricate interaction between several neurotransmitter systems, neural circuits and physiological processes. Pharmacological therapeutic approaches are commonly used in the treatment of insomnia to alleviate the acute and chronic symptoms of the sleep disorder. Multidisciplinary interventions for insomnia have been widely recognized as an effective and comprehensive approach to managing this sleep disorder. Conclusion: Understanding its complexities, from the underlying neurobiological mechanisms to the available therapeutic interventions, represents a fundamental step in addressing insomnia.
